Use of chemotherapy for patients with bone and soft-tissue sarcomas
For patients with bone sarcomas, chemotherapy has a proven role in the primary therapy of osteogenic sarcoma and Ewing sarcoma but no role for chondrosarcoma. Chemotherapy’s role is currently more limited for patients with soft-tissue sarcomas, as it is generally used to palliate metastatic disease in most subtypes of soft-tissue sarcoma and remains largely investigational in the treatment of operable disease. The chemotherapy regimens for musculoskeletal sarcomas often carry significant potential toxicities, so the efficacy of less intensive and less toxic regimens is a focus of ongoing research. Thrombin enhanced migration and MMPs expression of human chondrosarcoma cells involves PAR receptor signaling pathway
Thrombin is a multifunctional protease that can activate hemostasis and coagulation through the cleavage of fibrinogen to form fibrin clots. Thrombin also plays a crucial role in migration and metastasis of human cancer cells. However, the effect of thrombin on migration activity in human chondrosarcoma cells is mostly unknown. Here, we found that thrombin increased the migration and expression of matrix metalloproteinase (MMP)-2 and MMP-13 in human chondrosarcoma cells (JJ012 and SW1353 cells). Methylthioadenosine phosphorylase and activated insulin-like growth factor-1 receptor/insulin receptor: potential therapeutic targets in chordoma
Currently there is no effective chemotherapy for chordoma. Recent studies report co-expression of insulin-like growth factor-1 receptor (IGF1R) and its cognate ligand in chordoma, but it is unknown whether this receptor tyrosine kinase is activated in these tumours. Additionally, genetic studies have confirmed frequent deletions of chromosome 9p in chordomas, which encompasses the cyclin-dependent kinase inhibitor 2A (CDKN2A) locus. Synovial chondromatosis of spine: case report and review of the literature.

In this metaplastic process, islands of chondrocytes are clustered throughout the synovium, forming nodules that jut into the joint cavity. A peptidyl-glucosamine derivative affects IKKalpha kinase activity in human chondrocytes
Conclusions:
Our results demonstrate that glucosamine and its peptidyl-derivative can interfere with NF-kappaB signaling pathway by inhibiting IKKalpha activity in human chondrocytes. However, the mechanism of action of the two molecules is not completely overlapping. CXCR4/SDF1 mediate hypoxia induced chondrosarcoma cell invasion through ERK signaling and increased MMP1 expression

Chondrosarcoma cell invasion is increased by hypoxia induced expression of CXCR4 and MMP1 and is mediated by HIF-1a and ERK. BFPP, a phloroglucinol derivative, induces cell apoptosis in human chondrosarcoma cells through endoplasmic reticulum stress.
This study is the first to investigate the anticancer effects of the new phloroglucinol derivative (2,4-bis(2-fluorophenylacetyl)phloroglucinol; BFPP) in human chondrosarcoma cells. BFPP induced cell apoptosis in two human chondrosarcoma cell lines, JJ012 and SW1353 but not in primary chondrocytes. BFPP triggered endoplasmic reticulum (ER) stress, as indicated by changes in cytosol calcium levels, and increased glucose regulated protein 78 (GRP78) expression, but failed to show the same effects on GRP94 expression. BFPP also increased calpain expression and activity. Transfection of cells with GRP78 or calpain siRNA reduced BFPP-mediated cell apoptosis in JJ012 cells. No Recurrences in Selected Patients after Curettage with Cryotherapy for Grade I Chondrosarcomas.
CONCLUSIONS: The data confirm the appropriateness of conservative surgery for central low-grade chondrosarcomas of the proximal humerus and distal femur based on a combination of intralesional curettage and cryogenic parietal sterilization. Candidates for this approach should be chosen on the basis of the affected bone site, local extension staging, and clinicopathologic grading. We recommend supplementary internal fixation. Accelerated Ca2+ entry by membrane hyperpolarization due to Ca2+-activated K+ channel activation in response to histamine in chondrocytes.
In this study, the functional impact of Ca(2+) activated K(+) (K(Ca)) channels in the regulation of intracellular Ca(2+) concentration ([Ca(2+)](i)) in chondrocytes in response to histamine was examined using OUMS-27 cells, as a model of chondrocytes derived from human chondrosarcoma. Application of histamine induced a significant [Ca(2+)](i) rise and also membrane hyperpolarization, and both effects were mediated by the stimulation of H1 receptors. Laryngeal Chondrosarcoma: An Exceptional Localisation of a Not Unfrequent Bone Tumor
After osteosarcoma, chondrosarcoma is the second most common primary bone tumor accounting for 26% of all malignancies. In the laryngeal region however, chondrosarcomas are rather rare. Only 300 cases are reported in literature. Considering laryngeal chondrosarcoma, about 75% occur in the cricoid cartilage, whereas 20% occur in the thyroid cartilage.
