The transcription factor Lc-Maf participates in Col27a1 regulation during chondrocyte maturation.
Authors: Mayo JL, Holden DN, Barrow JR, Bridgewater LC
The transcription factor Lc-Maf, which is a splice variant of c-Maf, is expressed in cartilage undergoing endochondral ossification and participates in the regulation of type II collagen through a cartilage-specific Col2a1 enhancer element. Type XXVII and type XI collagens are also expressed in cartilage during endochondral ossification, and so enhancer/reporter assays were used to determine whether Lc-Maf could regulate cartilage-specific enhancers from the Col27a1 and Col11a2 genes. The Col27a1 enhancer was upregulated over 4-fold by Lc-Maf, while the Col11a2 enhancer was downregulated slightly. To confirm the results of these reporter assays, rat chondrosarcoma (RCS) cells were transiently transfected with an Lc-Maf expression p......

Circumferential Total Resection of Cervical Tumors: Report of Two Consecutive Cases and Technical Note.
Authors: Sımşek S, Belen D, Yığıtkanli K, Aciduman A, Bavbek M
To date, few studies have addressed the subaxial vertebrectomy technique and related anatomical landmarks in this method. Total spondylectomy is performed via piecemeal resection or en bloc removal in a one-stage procedure associated with stabilizing the spine and preserving neurological status. In this presentation, a circumferential total cervical tumor resection for subaxial cervical spine lesions was described. Two cases of subaxial cervical malignancy, one with primary C3 chondrosarcoma and the other with C4 lung adenosarcoma metastasis, were both treated by the anterior-posterior approach. The lesions could be removed macroscopically totally in both cases. The patients did well after surg......

Differentiating high-grade from low-grade chondrosarcoma with MR imaging
Abstract The purpose of the study was to evaluate the MR imaging features that differentiate between low-grade chondrosarcoma (LGCS)
and high-grade chondrosarcoma (HGCS) and to determine the most reliable predictors for differentiation. MR images of 42 pathologically
proven chondrosarcomas (28 LGCS and 14 HGCS) were retrospectively reviewed. There were 13 male and 29 female patients with
an age range of 23–72 years (average age 51 years). On MR images, signal intensity, specific morphological characteristics
including entrapped fat, internal lobular architecture, and outer lobular margin, soft tissue mass formation and contrast
enhancement pattern were analysed. MR imaging features used to identify LGCS and HGCS were compared using univariate analysis
and multi......

[Intracortical dedifferentiated chondrosarcoma.]
Authors: Thöm K, Bontikous S, Engelhardt M, Düring T, Schulte M
Chondrosarcoma is the second most frequent mesenchymal malignant tumour of the bone. Classification of this kind of tumour is made by clinical, radiological und pathological means. A case of an intracortical chondrosarcoma was first reported by Babinet et al. 2003 [2]. During the staging examination of a 59-year-old patient referred to our clinic because of a squamous cell carcinoma of the oropharynx, we also found a highly malignant intracortical dedifferentiated chondrosarcoma of the distal femur shaft. Due to the primary assumption of bone metastasis of the oropharynx tumour, marginal extralesional tumour resection was performed followed by composite osteosynthesis. Considerations on differential diagnosis and......

Development of chondrosarcoma animal models for assessment of adjuvant therapy
Chondrosarcoma is a primary cancer of bone causing significant morbidity due to local recurrence and limited treatment options. Relatively few chondrosarcoma animal models have been developed, and the only orthotopic model is technically demanding and has limited clinical relevance. The aim of this review is to assess the features of current animal chondrosarcoma models for the purpose of developing new models in which to test adjuvant chondrosarcoma therapy. The available literature on this topic was identified using the PubMed database, and then analysed for relevance to the human chondrosarcoma disease and feasibility in testing new therapeutic agents. Animal-derived chondrosarcoma models comprise predominantly allograft tumour transplanted into the rat (Swarm rat chondrosarcoma) or the......

The association between intracranial tumours and multiple dyschondroplasia (Ollier’s disease or Maffucci’s syndrome): do children and adults differ?
Abstract In Ollier’s disease (OD) and Maffucci’s syndrome (MS), there is deforming dysplasia of cartilage, primarily but not exclusively
involving the metaphyses and diaphyses of long bones. In a minority of patients with either of these rare syndromes, dysplasia
can lead to sarcomatous degeneration, producing chondrosarcomas. There also appears to be an association with other neoplasms,
which can include intracranial tumours. The primary objective of the current paper was to compare children/adolescents who
have either OD or MS and an intracranial malignancy with their adult counterparts. All relevant cases in the medical literature
were identified by electronically searching PubMed, SciSearch, Scientific Commons, Springer Link, and Google. Translate DotNet

Antitumorigenic Effect of Brain Proline Rich Polypeptide-1 in Human Chondrosarcoma.
Authors: Galoian K, Scully S, McNamara G, Flynn P, Galoyan A
Proline rich polypeptide (PRP-1) produced by neurosecretory cells of the hypothalamus is one of the fragments of neurophysin-vasopressin-associated glycoprotein. The primary structure of the neuropeptide PRP-1 isolated from neurosecretory granules of bovine neurohypophysis. We investigated PRP-1 action on chondrosarcoma, the second most common malignancy in bone, which primarily affects the cartilage cells. This deadly disease does not have any effective treatment. Earlier we demonstrated MYC oncogene inactivating effect by 1 mug/ml concentration brain PRP-1 In the present study we observed reduced viable sarcoma JJ012 cell numbers in comparison with control (89% growth inhibition) when treated with low concentrations of PRP-......

Osteopontin increases migration and MMP-9 up-regulation via [alpha]v[beta]3 integrin, FAK, ERK, and NF-[kappa]B-dependent pathway in human chondrosarcoma cells
Tumor malignancy is associated with several features such as proliferation ability and frequency of metastasis. Osteopontin (OPN), which abundantly expressed in bone matrix, is involved in cell adhesion, migration, invasion and proliferation via interaction with its receptor, that is, [alpha]v[beta]3 integrin. However, the effect of OPN on migration activity in human chondrosarcoma cells is mostly unknown. Here we found that OPN increased the migration and expression of matrix metalloproteinase (MMP)-9 in human chondrosarcoma cells (JJ012 cells). RGD peptide, [alpha]v[beta]3 monoclonal antibody and MAPK kinase (MEK) inhibitors (PD98059 and U0126) but not RAD peptide inhibited the OPN-induced increase of the migration and MMP-9 up-regulation of chondrosarcoma cells. OPN stimulation increase......

[RSNA Education Exhibits] Imaging Appearances of the Sternum and Sternoclavicular Joints
The sternum and sternoclavicular joints—critical structures of the anterior chest wall—may be affected by various anatomic anomalies and pathologic processes, some of which require treatment. Pectus excavatum and pectus carinatum are common congenital anomalies that are usually benign but may warrant surgical treatment if they cause compression of vital internal structures. By contrast, developmental variants such as the sternal foramen are asymptomatic and do not require further evaluation or treatment. Arthritides of the sternoclavicular joint (osteoarthritis, septic arthritis, and seronegative arthropathies) are common and must be differentiated before an appropriate management method can be selected. The recognition of complications of sternotomy (eg, sternal dehiscence, se......

siRNA-based targeting of antiapoptotic genes can reverse chemoresistance in P-glycoprotein expressing chondrosarcoma cells
Conclusions:
These findings suggest that antiapoptotic proteins play a significant role in the chemoresistance of chondrosarcoma cells independent of P-glycoprotein. Based on the results, a new siRNA-based therapeutic strategy targeting antiapoptotic genes can be designed to overcome the chemoresistance of chondrosarcomas which is often conferred by P-glycoprotein. [CT and MRI of intrinsic space-occupying lesions of the bony skull base.]
This article gives an overview of the appearance of the most common primary bony skull base masses in CT and MRI. From the authors' point of view these are fibrous dysplasia, chordomas, chondrosarcomas, Langerhans cell histiocytosis and multiple myelomas, which must be differentiated from pseudolesions. The possibilities of CT and MRI in making a specific diagnosis, differential diagnosis and the kind of making the final diagnosis are described.

“Scrotal Pouch”—Scrotal Herniation of Bladder Secondary to Extensive Bone Resection Due to Chondrosarcoma: A Simple and Effective Surgical Treatment of Urinary Obstruction
A patient developed a scrotal hernia of the bladder 6 years after resection of the pubic ramus for treatment of a chondrosarcoma. Because reconstruction of the pelvis was impossible, an alternative surgical solution was needed. A continent vesicocutaneous stoma with a full-thickness bladder tube was created. The hernia itself was not repaired, leaving the bladder and bowel loops in the scrotum. Nine years after surgery, the patient continued to perform clean intermittent catheterization. The patient had not experienced any strictures or infections. The demonstrated technique appears to be a feasible option when hernia repair is impossible. (Source: Urology)...

Proton therapy for tumors of the skull base
Abstract Charged particle beams are ideal for treating skull base and cervical spine tumors: dose can be focused in the target, while
achieving significant sparing of the brain, brain stem, cervical cord, and optic nerves and chiasm. For skull base tumors,
10-year local control rates with combined proton-photon therapy are highest for chondrosarcomas, intermediate for male chordomas,
and lowest for female chordomas (94%, 65%, and 42%, respectively). For cervical spine tumors, 10-year local control rates
are not significantly different for chordomas and chondrosarcomas (54% and 48%, respectively), nor is there any difference
in local control between males and females. Observed treatment-related morbidity has been judged acceptable, in view of the
major morbidity and mortali......

Extended endoscopic approaches for midline skull-base lesions
Abstract The endoscopic transsphenoidal approach has been reported in the literature as a useful tool to treat sellar and parasellar
lesions. The endoscope permits a panoramic view instead of the narrow microscopic view, and it allows the inspection and removal
of the lesions of sellar, parasellar, and suprasellar compartments by angled-lens endoscopes. On the basis of the experience
gained with the use of the endoscope, we have performed extended endoscopic endonasal transsphenoidal approach in 13 of 200
(total endoscopic transphenoidal approaches since September 1997) patients for the last 5 years. Extended endoscopic transsphenoidal
approach was performed for three patients with pituitary adenoma, two patients with craniopharyngioma, one patient with metastatic

Chondrosarcoma in the United States (1973 to 2003): An Analysis of 2890 Cases from the SEER Database
Conclusions: Only grade and stage are independent prognostic factors for survival in cases of chondrosarcoma. Current treatment algorithms have not improved the survival rates of patients with chondrosarcoma over the past thirty years. Routine patient surveillance following treatment should be extended to ten years of follow-up.
